Short Description

The CytoFLEX LX is a high-end flow cytometer from Beckman Coulter, equipped with six lasers (355, 405, 488, 561, 638 und 808 nm) and 21 fluorescence detectors. Two further detectors for FSC and SSC signals are available. The SSC signal can be collected either from the blue or from the violet laser. The CytoFLEX LX is equipped with Avalanche photodiode detectors instead of the more conventional photomultiplier tubes. According to Beckman Coulter, these detectors have an increased sensitivity for fluorescent light from the infrared spectrum.

The following filters and detectors are assigned to the different lasers: UV-Laser (355nm): three detectors with 405/30, 525/40 and 675/30 bandpass-filters; violet laser (405nm): five detectors with 450/45, 525/40, 610/20, 660/10 and 763/43 bandpass-filters; blue laser (488nm): three detectors with 525/40, 610/20 and 690/50 bandpass-filters; yellow-green laser (561 nm): five detectors with 585/42, 610/20, 675/30, 710/50 and 763/43 bandpass-Filters; red laser (638 nm): three detectors with 660/10, 712/25 and 763/43 bandpass-filters; infrared laser (808 nm): two detectors with 840/20 und 885/40 bandpass-filters. Samples can be acquired out of 12 x 75 mm tubes as well as 2 or 1.5 ml Microfuge-tubes. A plate loader is NOT part of the system. The system can handle up to 30,000 events per second; the flow rate can be varied between 10 and 240µl per minute. The system is operated by the CytExpert software of Beckman Coulter, acquired data is saved in the FCS 3.0 format, which allows further data processing with other flow cytometry software packages.

Methods & Expertise for Research Infrastructure

The CytoFLEX LX is located at the Institute of Immunology at the University of Veterinary Medicine Vienna. The members of the Institute of Immunology work mainly on the immune system of farm and pet animals. The main focus is on the pig, to a lesser extent also chicken, turkey, cattle, dog and cat are investigated. Phenotyping of immune cells by polychromatic flow cytometry is a major focus of the work. In addition, several functional analyses of immune cells are performed by flow cytometry; among them are proliferation, cytokine production or detection of phosphorylated signal transduction molecules. Members of the Institute have a high expertise in the establishment of multi-color staining panles with leukocytes from the aforementioned species.
