Open for Collaboration

Short Description

The Core Facility Cytometry offers a wide range of highly specialized services in the field of cellular analysis and sorting for biomedical research. Equipped with state-of-the-art flow cytometry and cell sorting instruments, it supports researchers in the investigation and characterization of cell populations, immunophenotyping, cell cycle analysis and cell function studies. In addition to the provision and maintenance of equipment and various software, the Core Facility Cytometry also offers training and highly qualified support in the areas of sample preparation, project planning, analysis and interpretation of data.

The Core Facility Cytometry is located at the Center for Medical Research (ZMF) of the Johannes Kepler University Linz. A total of 5 high-end flow cytometers are available for cell characterization and cell sorting. The devices cover the entire spectrum from multiparameter and spectral flow cytometry to sterile cell sorting of up to 4 subpopulations with up to 18 parameters.

Depending on the problem and sample characteristics, each device serves a different measurement purpose, e.g. for high-throughput measurements, measurement of different cell sizes and particles, for cells with different autofluorescence, for complex multi-color analyses or for sorting different subpopulations or single cell sorting.

Research Services

The aim of the Core Facility Cytometry is to offer scientists access to state-of-the-art flow cytometry devices:

4 analyzers (CytoFLEX V5-R3-B5, DsFLEX V5-R3-B5, Northern Lights V16-B14-R8, Guava®easyCyte™12HT)

Our Core Facility is equipped with three state-of-the-art flow cytometers, each with 3 lasers (405, 488, 640 excitation wavelength) up to 13 fluorescence channels and a spectro-cytometer up to 38 fluorescence channels.

The spectral detection offers advantages such as the simultaneous use of dyes such as BV421, Super Bright 436 and eFluor® 450, APC and Alexa Fluor® 647, the optimized detection of weak fluorescent labels through autofluorescence correction and the combination of two side scatters (488/405 SSC).

After successfully completing the necessary training and after approval by the staff of the Core Facility, these devices are also available to scientists for independent use.

2 sorters (Biorad S3e Benchtop Sorter and BD FACS Aria Fusion High-End Sorter)

BioRad's S3e Benchtop Sorter with few fluorescence detectors in combination with a sophisticated, user-friendly technology simplifies cell sorting, enables simultaneous separation of up to two subpopulations and deposition of single cells in different formats.

The BD FACS Aria Fusion with five lasers (355nm, 405nm, 488nm, 561nm, 640nm) and 18 fluorescence channels enables complex high-speed cell sorting, high-purity sorting, storage in different formats and simultaneous separation of up to four subpopulations.

The cell sorters are operated by the staff of the Core Facility. However, it is possible for the Core Facility to grant individual scientists with sufficient experience and who have completed the necessary training independent authorization to use the equipment.
