Short Description

The PerkinElmer Operetta CLS is an innovative high-content/high-throughput analysis system with a confocal option. The device is specialized in obtaining more physiologically relevant information through complex image analyses at high throughput.

The unique features of the PerkinElmer Operetta CLS™ are diverse and include, among other things, an excitation spectrum equipped with 8 LEDs covering a range from UV to near-infrared, a 4-position emission filter wheel, and three water-immersion objectives (20x, NA 1.0; 40x, NA 1.1; 63x, NA 1.15), which enable higher Z-resolution for 3D analyses as well as shorter measurement times.

Furthermore, the system is controlled by Harmony, a user-friendly software platform, and plate data can be automatically transferred to the PerkinElmer Columbus image storage and analysis system.

Methods & Expertise for Research Infrastructure

Using the PerkinElmer Operetta CLS, we perform high-content, high-throughput imaging and quantitative analysis of complex 3D tissues and organoids. The system’s confocal capabilities and water-immersion objectives enable high-resolution 3D reconstruction and detailed phenotypic characterization. We conduct automated test series and compound screenings in multiwell formats, as well as functional live-cell analyses, including mucociliary clearance assays and dynamic cellular response measurements under physiologically relevant conditions.
