JKU - Johannes Kepler University Linz

Linz

Open for Collaboration

Short Description

Imaging flow cytometer with high-resolution camera detection, enables simultaneous recording of phase contrast images and (up to 9) fluorescence channels of cells / particles in suspension. Four lasers (405 nm, 488 nm, 561 nm, 632 nm) for fluorescence excitation, white light LED for phase contrast and IR laser for side scatter detection. Four objectives (20x, 40x and 60x) for different cell / particle sizes.

Methods & Expertise for Research Infrastructure

The high-performance imaging cytometer enables the simultaneous recording of white light and fluorescence images with high sample throughput and thus significantly expands the application possibilities of flow cytometry. Measurements are possible with living or fixed cells or other particles in suspension. The sample preparation does not differ from conventional flow cytometry; for the most part, the equivalent fluorescent dyes and antibodies can be used. The Ideas 6.2 evaluation software enables advanced image analysis (predefined and user-defined masks, colocalization, advanced parameters such as anisotropy, aspect ratio, etc.), quantitative evaluation and automated fluorescence compensation.
