Methods & Expertise for Research Infrastructure

CytoFLEX LX is a classical (detector-based) flow cytometer. The device is equipped with 6 lasers (355nm, 405nm, 488 nm, 561 nm, 638 nm and 808 nm) and 21 avalanche photodiode (APD) fluorescence detectors. The APD detector gains are calibrated for a linear response meaning that measured fluorescence signal intensities are linear to the detector gain setting. This enables "gain-independent" compensation, where the compensation matrix obtained at one gain setting can be used following re-calculation by the software for experiments at different gain settings. The software automatically recalculates spillover values in real time as the gains are adjusted.
Measurements on this device can only be made from tubes (Eppendorf vials, 5 ml FACS tubes). Cell concentration can be recorded without additional counterbeads.
