Short Description

The MACSima Imaging Platform is a fully automated fluorescence microscope that enables imaging of 200+ proteins on a single sample (FFPE, fresh frozen, or cell culture), thereby providing access to spatial proteomics. The system performs cyclic staining, imaging, and stain removal fully automatically. Multi-omics analysis is possible through imaging of up to 27 RNAs or in combination with other technologies.

Methods & Expertise for Research Infrastructure

The microscope is suitable for capturing up to 200-plex immunofluorescence markers on the same sample. One staining cycle includes staining, image acquisition, and signal removal. Three colour channels are available per cycle, up to 65 cycles can be included in a run. Signal removal is performed enzymatically or via photobleaching.The supervisors have expertise in panel design, experiment optimization, troubleshooting, experiment execution, and data analysis using the associated MACSiQ View software. In data analysis, differentially expressed cells can be identified, quantified, and localized using FACS-like gating.

Possible research questions: How does the localization and percentage of different cell populations differ in healthy versus diseased tissue? Are different tumor cell subtypes concentrated in distinct niches, and which types of immune cells are located nearby?
